200 Optimised 3D skin models incorporating microbiome and host immune responses

Commensal microbes and humans have coevolved toward a symbiotic relationship. In healthy individuals, the human microbiome has vital role in development of innate adaptive immune system, inducing protective regulatory immunity, system maintains an optimal balance microbes. Several acute chronic skin conditions are result imbalances host-microbiome relationship accompanied by epidermal barrier disruption. This research aims to develop three-dimensional (3D) vitro models integrating microbiology cells simulate this complex effects on health. 3D models, created using immortalised keratinocyte cell line NTERTs seeded collagen matrices embedded with dermal fibroblasts, were colonised for 72 hrs singular or combination inoculums 5 prominent species: Staphylococcus epidermidis, capitis, Cutibacterium acnes, Malassezia restricta globosa. Colonisation showed differences K14, K16, K10, Ki67 involucrin expression. Epidermal thinning was observed inoculations S. C. acnes M. however, restoration thickness compared control models.
